PIGP also known as phosphatidylinositol glycan anchor biosynthesis class P acts mechanically in the early step of glycosylphosphatidylinositol (GPI) anchor biosynthesis. This protein plays an essential role in the transfer of N-acetylglucosamine (GlcNAc) from UDP-GlcNAc to phosphatidylinositol (PI) on the cytoplasmic side of the endoplasmic reticulum. PIGP has an estimated molecular mass of around 10 kDa. The gene encoding PIGP expresses predominantly in several tissues especially in the brain and other neural tissues.
Biological function summary
PIGP functions as an important component of the GPI transamidase complex—an important part of the GPI anchor biosynthesis process. This complex includes several other proteins such as PIGQ PIGT PIGK and GPAA1 which coordinate closely for GPI anchor synthesis. GPI anchors attach proteins to the cell membrane affecting their localization and function. Therefore PIGP contributes significantly to diverse cellular mechanisms.
Pathways
PIGP participates integrally in the GPI-anchor biosynthesis pathway and the inositol phosphate metabolism pathway. It closely interacts with other proteins like PIGA and PIGC that are involved in the same biosynthetic pathway. These interactions ensure the proper assembly and function of GPI-anchored proteins which play substantial roles in signal transduction and cell adhesion processes.
PIGP has associations with inherited conditions such as Paroxysmal Nocturnal Hemoglobinuria (PNH) and Down syndrome-related mental disorders. In PNH aberrations in the GPI-anchor biosynthesis disrupt cell membrane stability often involving proteins like PIGA. Meanwhile specific mutations within this pathway may link to intellectual disabilities seen in Down syndrome illustrating the clinical significance of PIGP and related proteins in human health.
