FIAT also known as Factor Inhibiting ATF4-mediated Transcription is a transcriptional regulator with a molecular weight of approximately 37 kDa. You can find it expressed in various tissues but it is especially prevalent in bone tissue. Mechanically FIAT interacts with ATF4 inhibiting its transcriptional activity. FIAT binds to the bZip region of ATF4 preventing ATF4 from regulating genes that are critical for osteoblast differentiation and function.
Biological function summary
FIAT plays an important role in bone development and homeostasis. It is not known to be part of a large protein complex but it functions by altering transcriptional outputs. FIAT limits ATF4's ability to activate osteoblast-specific target genes consequently influencing bone formation and remodeling. By doing this FIAT helps to maintain proper bone density and strength balancing the activities of osteoblasts and osteoclasts.
Pathways
Research indicates that FIAT is involved in the osteoblastic bone formation pathway. It regulates this pathway through its interaction with ATF4 which is an important player in translating extracellular signals to gene expression responses. FIAT also intersects with the BMP (Bone Morphogenetic Protein) pathway influencing BMP target genes and interacting with other transcription factors like Runx2.
FIAT links to conditions affecting bone health such as osteoporosis. Due to its inhibition of ATF4 FIAT indirectly contributes to osteoporosis by reducing osteoblast activity leading to decreased bone density. FIAT's relationship with ATF4 connects it to the regulation of genes altered in osteoporotic bones.
