MPP8 also known as M-phase phosphoprotein 8 or MPG functions mechanically as a chromodomain-containing protein involved in chromatin organization. It has a mass of approximately 92 kDa. MPP8 is expressed in several human tissues including the liver lung and brain. As an epigenetic regulator MPP8 plays a significant role in regulating gene expression through its interactions with chromatin.
Biological function summary
Within the cellular environment MPP8 participates in maintaining heterochromatin structure which is essential for proper chromosomal behavior. It is a component of the HUSH complex (human silencing hub) which is important for transcriptional repression. This protein complex involves other proteins like TASOR and PERIPHILIN collaborating to guide transcriptional silencing to specific genomic regions. MPP8's chromodomain allows it to recognize and bind to methylated histones further aiding its role in chromatin modification.
Pathways
MPP8 participates in epigenetic regulation and gene silencing pathways. It is notably involved in the silencing of retrotransposon elements through the HUSH complex. Proteins like TASOR enhance the silencing function of MPP8 in these pathways. MPP8 ensures proper chromatin state maintenance which influences downstream gene transcription processes.
