APOBEC3F also known as APOBEC-related complex 3F is a member of the APOBEC family of cytidine deaminases. This protein functions by deaminating cytosine bases in single-stranded DNA leading to the conversion of cytosine to uracil. APOBEC3F has a molecular mass of approximately 46 kDa. It is primarily expressed in human immune cells including T lymphocytes and certain monocytes where it performs its enzymatic activity in response to intracellular threats.
Biological function summary
APOBEC3F acts as an important component in the cellular defense against retroviruses such as HIV. The protein incorporates into HIV virions and induces hypermutation when the virus attempts to integrate into the host genome impairing viral replication. APOBEC3F forms a complex with other APOBEC3 family members enhancing its antiviral activity. This collaboration heightens its capacity to neutralize and control viral infections within the host.
Pathways
APOBEC3F plays a significant role in the innate immune response. It engages with the HIV-1 replication pathway inhibiting the action of viral proteins like Vif which aims to counteract APOBEC3F's defensive effects. The protein also connects with pathways involving other APOBEC3 enzymes contributing to a broader cytidine deamination effort to prevent viral propagation.
The activity of APOBEC3F holds relevance to HIV/AIDS. Its interaction with the HIV-1 protein Vif can determine susceptibility to viral replication and progression of the disease. Additionally aberrant activity of APOBEC3F may link to mutagenesis in cancer as excessive or uncontrolled deamination may induce mutations in oncogenes or tumor suppressor genes. The study of APOBEC3F along with related proteins such as APOBEC3G continues to shed light on its contribution to both viral immunity and oncogenic processes.
