The protein Neuropathy Target Esterase (NTE) also known as PNPLA6 is an important enzyme that catalyzes the hydrolysis of specific phospholipids. This enzyme is approximately 150 kDa in mass. It is widely expressed in neuronal tissues but is found in various other tissues such as liver and kidneys as well. By removing specific side groups from phospholipids NTE regulates lipid metabolism and membrane dynamics which is essential for maintaining cellular function and integrity.
Biological function summary
The Neuropathy Target Esterase functions in the detoxification of neuropathic organophosphates and plays critical roles in neural development and maintenance. NTE is part of a complex involved in the regulation of membrane phospholipid homeostasis. This complex acts to monitor and maintain the composition of membranes which is important for cell signaling and adhesion processes. The absence or malfunction of NTE can lead to disruptions in these processes indicating its importance in normal neuronal activity.
Pathways
NTE participates in phospholipid biosynthesis and degradation pathways. It interacts with proteins involved in membrane trafficking and signaling such as phospholipase D. These pathways are critical for lipid signaling and membrane dynamics linking NTE to key processes in cellular communication and response. NTE's association with these pathways emphasizes its role in maintaining lipid balance and cellular communication through its enzymatic activity.
NTE mutations or dysfunctions connect to complex conditions such as hereditary spastic paraplegia and motor neuron disease. These mutations can disrupt its known interactions with proteins like REEP1 and spastin within the nerve cells. Such conditions often involve the degeneration of motor neurons leading to progressive motor dysfunction. Understanding NTE's role in these diseases may provide insights for therapeutic interventions targeting these neurodegenerative disorders.
