How the assay works
Human Prekallikrein 1B CatchPoint SimpleStep ELISA employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. CatchPoint HRP Development Solution containing the Stoplight Red Substrate is added. During incubation, the substrate is catalyzed by HRP generating a fluorescent product. Signal is generated proportionally to the amount of bound analyte and the intensity is measured in a fluorescence plater reader at 530/570/590 nm Excitation/Cutoff/Emission. Prekallikrein 1B (KLKB1) is a 88 kDa serine protease involved in blood coagulation, kinin generation, fibrinolysis and inflammation. Mature KLKB1 primarily circulates as a complex with high-molecular weight kinnogen, where it releases kinins from kininogens during the inflammation and vasodilation response.
This kit detects both prekallikrein and plasma kallikrein.

Plasma Kallikrein 1B also known as PKK or KLKB1 is a serine protease enzyme with an approximate mass of 88 kDa. It is important for the blood coagulation process. This protein is mainly expressed in the liver and circulates in the blood as the inactive precursor prekallikrein. Through cleavage prekallikrein converts into its active form plasma kallikrein. Plasma kallikrein exhibits various proteolytic activities influencing multiple substrates involved in hemostatic and inflammatory responses.
Biological function summary
Plasma Kallikrein 1B plays significant roles in the kinin-kallikrein system. Within this system it cleaves high-molecular-weight kininogen to release bradykinin a potent vasodilator. This enzyme is not part of a larger protein complex but operates independently to modulate various physiological responses. Plasma kallikrein's activity influences processes like pain vasodilation and inflammation making it important in homeostasis and acute-phase responses.
Pathways
Plasma Kallikrein 1B participates actively in the kallikrein-kinin and the contact activation pathways. In the kallikrein-kinin system it works alongside high-molecular-weight kininogen and factor XII which plays an important role in the intrinsic pathway of coagulation.
