The cxcr4b protein also known as C-X-C motif chemokine receptor 4b is a G-protein coupled receptor with a mass of approximately 40-50 kDa. It is expressed in various tissues most notably in the hematopoietic and nervous systems. The receptor is located on the cell surface and plays a critical role in mediating cell signaling processes. It is especially known for its function in regulating the trafficking of cells in response to its ligand the stromal-derived factor 1 (SDF-1) also known as CXCL12.
Biological function summary
Cxcr4b influences cell migration survival and development. It is part of the CXCR4/CXCL12 signaling axis which involves a complex interaction between the receptor and its chemokine ligand. This axis is responsible for guiding the movement of cells especially during embryonic development and immune surveillance. The receptor also supports the proper formation of blood vessels and maintenance of various stem cell niches within tissues.
Pathways
Cxcr4b holds a significant position in both the chemokine-mediated signaling pathway and the Jak-STAT signaling pathway. Within these pathways cxcr4b plays an integral role by interacting with other proteins such as CXCL12 and Jak kinases facilitating downstream activation that influences cell behavior. These interactions lead to effects on effector functions like cell proliferation and gene expression.
Cxcr4b is closely associated with cancer progression and HIV infection. Enhanced expression of cxcr4b often correlates with tumor growth and metastasis making it an area of interest in oncology. In the context of HIV cxcr4b acts as a coreceptor that allows the virus entry into host cells working together with CD4 another protein critical in the viral pathogenicity. Understanding cxcr4b's interactions in these diseases helps in developing targeted therapies for better disease outcomes.
