Keratocan also known as Kera or Keratan sulfate proteoglycan plays a structural role in connective tissues. This small leucine-rich proteoglycan (SLRP) has an approximate mass of 37 kDa. It is expressed mainly in the cornea but also found in cartilage and other connective tissues. The arrangement of keratocan contributes significantly to the transparency and biomechanical properties of the cornea interacting with collagen fibrils.
Biological function summary
Within the extracellular matrix keratocan maintains corneal clarity and supports tissue organization. It is part of the corneal proteoglycan complex which includes lumican and decorin. These SLRPs collectively regulate collagen fibril assembly and spacing ensuring proper light refraction through the cornea. Keratocan’s presence is also indicative of its role in corneal morphogenesis and repair assisting in wound healing processes.
Pathways
Keratocan interacts with the TGF-β and BMP signaling pathways contributing to matrix assembly and cellular growth. These pathways involve other proteins like fibromodulin and biglycan sharing functions in collagen assembly and tissue repair.
