ZDHHC4 also known as DC1 is a palmitoyltransferase enzyme with a mass of approximately 35 kDa. It plays an important role in attaching palmitate a fatty acid to cysteine residues on target proteins which is an important post-translational modification. This modification helps in the trafficking stability and function of proteins within a cell. ZDHHC4 is expressed in various tissues including the brain and liver influencing diverse biological processes and cellular functions.
Biological function summary
This palmitoyltransferase influences protein-protein interactions by adding lipid moieties to key signaling molecules. It is part of the ZDHHC family of proteins which collaborate to modify a variety of substrates important for cellular signaling pathways. By palmitoylating proteins ZDHHC4 can regulate pathways involving signal transduction and vesicular trafficking affecting how signals are transmitted inside the cell and how proteins are transported to different cellular compartments.
Pathways
Research shows this enzyme acts within the signaling pathways such as the Wnt signaling and Ras signaling. In the Wnt pathway ZDHHC4's palmitoylation activity helps control the stability and localization of proteins important for signal propagation. Within the Ras signaling pathway it modifies Ras proteins which are related to cell growth and survival. These associations highlight its role in influencing cell behavior through complex signaling networks.
Mutations or dysregulation of ZDHHC4 have links to neurodegenerative diseases and cancer. For instance its activity impacts protein interactions that when impaired can contribute to Alzheimer's disease. In cancer ZDHHC4's altered palmitoylation activity can affect oncogenic signaling cascades contributing to tumor progression. It shows interaction with proteins such as beta-catenin in cancer and Beta-amyloid precursor in Alzheimer's connecting ZDHHC4 to these diseases at a molecular level.
