HEXO also known as Hexokinase is an enzyme with several isoforms expressed in different tissues. It plays an important role in the initial step of glycolysis by phosphorylating glucose to form glucose-6-phosphate. The mass of HEXO varies depending on the isoform; for example Hexokinase I typically has a mass around 102-104 kDa. HEXO is expressed in the cytoplasm of cells but specific isoforms like Hexokinase II are found on the outer membrane of mitochondria. This mitochondrial association is important for its regulatory function and activity in tissues such as muscle and brain.
Biological function summary
HEXO is significant in energy metabolism. It converts glucose into glucose-6-phosphate setting off the glycolytic pathway and linking to other pathways such as the pentose phosphate pathway. HEXO often functions as part of a larger enzyme complex within the cell collaborating with other glycolytic enzymes to efficiently metabolize glucose. Its activity influences cellular energy homeostasis and biosynthetic processes impacting cellular growth and division.
Pathways
HEXO is a central enzyme in carbohydrate metabolism and plays an important role in the glycolysis and pentose phosphate pathways. In glycolysis HEXO catalyzes the first committed step which is important for the production of ATP and metabolic intermediates. In the pentose phosphate pathway it provides glucose-6-phosphate as a substrate which is essential for the generation of NADPH and ribose-5-phosphate. Proteins like Glucose-6-phosphate dehydrogenase (G6PD) work with HEXO in the pentose phosphate pathway showcasing a collaborative role in oxidative stress response and anaplerotic reactions.
Impaired hexokinase function can be linked to conditions such as diabetes and cancer. These disorders exhibit alterations in glucose metabolism where reduced or aberrant activity of HEXO affects cellular energy balance and proliferation. For instance decreased HEXO activity relates to insulin resistance in diabetes impacting glucose uptake and metabolism. In cancer the unique association of certain hexokinase isoforms with mitochondria influences apoptosis resistance often involving proteins like VDAC (Voltage-dependent anion channel) which HEXO interacts with contributing to altered metabolic phenotypes in cancer cells.
