TBC1D20 also known as TBC domain family member 20 is a Rab GTPase-activating protein with an approximate mass of 45 kDa. This protein is primarily expressed in the endoplasmic reticulum and is involved in the regulation of Rab GTPases which are critical for intracellular membrane trafficking. TBC1D20 interacts specifically with Rab1 and Rab2 playing a role in the control of vesicle formation trafficking and fusion processes.
Biological function summary
TBC1D20 influences cellular functions by acting as a regulator of autophagy and endocytosis processes. It participates in the modulation of endoplasmic reticulum morphology influencing its shape and function. TBC1D20 does not typically form part of any larger complex but acts through its interactions with Rab proteins and other associated proteins involved in vesicle transport. Its activity is vital for maintaining proper cellular homeostasis by ensuring efficient cargo delivery.
Pathways
TBC1D20 plays a part in the endocytic pathway and autophagic processes important for cellular nutrient regulation and waste management. It interacts with proteins such as LC3 which are instrumental in autophagosome formation. TBC1D20 ensures the timely deactivation of Rab proteins coordinating with other GTPase-activating proteins to enforce proper vesicular trafficking along these pathways.
TBC1D20's dysfunction is linked to conditions like Charcot-Marie-Tooth disease and some neurodegenerative disorders. The protein's role in membrane trafficking and autophagy ties it to the etiology of these diseases with disruptions in these pathways leading to pathological manifestations. TBC1D20 is also connected to proteins such as MFN2 which is involved in mitochondrial dynamics highlighting its broader impact on cellular function through pathway disruption.
