PIGV also known as phosphatidylinositol glycan anchor biosynthesis class V is an enzyme that plays an essential role in the biosynthesis of glycosylphosphatidylinositol (GPI) anchors. These anchors attach certain types of proteins to cell membranes. PIGV has a molecular mass of approximately 57 kDa. The gene encoding PIGV primarily expresses in various tissues including the liver and brain suggesting it plays distinct roles across different biological systems.
Biological function summary
The protein PIGV participates in the GPI anchor biosynthesis pathway which is vital for the post-translational modification of proteins. This protein acts as part of a GPI-mannosyltransferase II complex within the endoplasmic reticulum. The complex adds mannose to the growing GPI anchor an important step for anchoring proteins to the cell surface. Such anchored proteins are often involved in cell signaling immune response and interactions between cells.
Pathways
The PIGV protein functions within the broader GPI-anchor biosynthesis pathway. It contributes directly to the synthesis of the GPI-anchor component critical for cellular processes involving surface proteins. The PIGV enzyme operates alongside PIGO and other GPI-related proteins in this pathway ensuring the precise assembly of the GPI-anchor which is necessary for the anchoring function.
PIGV mutations can lead to Hyperphosphatasia with mental retardation syndrome type 1 (HPMRS1) a condition that features intellectual disability along with elevated levels of serum alkaline phosphatase. This syndrome associates with defects in GPI-anchor biosynthesis linking PIGV closely to the pathogenic process. Additionally other proteins within the GPI biosynthesis pathway such as PIGM may share pathogenic relationships and influence the same syndrome through similar mechanisms highlighting PIGV's significance in GPI-related disorders.
