SLC12A6 also known as KCC3 is a potassium-chloride cotransporter involved in regulating ion homeostasis. The protein is big weighing approximately 135 kDa. It moves potassium and chloride ions across cell membranes which affects osmotic balance and cell volume. KCC3 is found widely in the human body with higher expression in the nervous system kidneys and heart. This wide expression suggests its important role in maintaining balance of ions in various tissues.
Biological function summary
KCC3 impacts cellular processes by being an important part of regulating salt balance within cells. It belongs to the cation-chloride cotransporter family and plays an important role in cell volume regulation. KCC3 does not usually function as part of a larger protein complex but acts as a standalone transporter. It helps stabilize cell environment which is necessary for correct cellular signaling and function.
Pathways
KCC3 is integral to maintaining the proper function of the WNK-SPAK/OSR1 kinase pathway. This pathway regulates ion transport and adjusts cell volume after osmotic stress. KCC3 interacts with proteins from this pathway like WNK kinases and SPAK which phosphorylate KCC3 to control its activity. Mutations in KCC3 link to Hereditary Motor and Sensory Neuropathy with Agenesis of the Corpus Callosum (HMSN/ACC).
