Roquin-2 also known as RC3H2 is a protein recognized for its role in post-transcriptional regulation. The protein has a mass of approximately 125 kilodaltons. You can find Roquin-2 primarily expressed in immune cells such as T-cells and B-cells but it is also present in other tissues. It contains a RING finger domain that allows it to function as an E3 ubiquitin ligase. This domain plays an important role in protein-protein interactions and tagging other proteins for degradation.
Biological function summary
Roquin-2 regulates the stability and translation of several mRNAs involved in inflammatory responses. Acting as a post-transcriptional modulator Roquin-2 binds to specific sequences in the 3' untranslated region (UTR) of target mRNAs affecting their degradation or translation. Roquin-2 is part of a larger complex that includes other regulatory proteins allowing for precise control over mRNA fate. This control impacts various immune functions which is vital for maintaining immune homeostasis.
Pathways
Research has shown that Roquin-2 integrates into the immune response pathways. It has significant involvement in the NF-κB signaling pathway where it modulates the production of cytokines. Roquin-2 also interacts with the TTP protein which is another post-transcriptional regulator highlighting its role in inflammation-related pathways. These interactions influence gene expression patterns that are necessary for appropriate immune functions.
Current studies indicate Roquin-2 has implications in autoimmune diseases such as systemic lupus erythematosus. Alterations in Roquin-2 function can lead to dysregulated immune responses contributing to such disorders. Roquin-2 is connected to proteins involved in autoimmune pathways including ICOS which plays a role in T-cell activation. Understanding how Roquin-2 controls these pathways could offer insights into therapeutic approaches for autoimmune conditions.
