Spermidine synthase also known as SPDSY or SRM catalyzes the production of spermidine from putrescine. This enzymatic reaction involves decarboxylated S-adenosylmethionine and is an essential step in polyamine synthesis. Spermidine synthase is a well-characterized enzyme with a molecular mass of approximately 35 kDa. The enzyme is predominantly expressed in tissues with high cellular turnover and proliferation such as the liver kidney and testis.
Biological function summary
Spermidine synthase plays a critical role in the synthesis of polyamines such as spermine which are vital for cell growth and differentiation. Polyamines stabilize DNA structure modulate ion channels and aid in gene expression. Spermidine synthase operates as a standalone enzyme and is not part of a multi-protein complex. Its expression levels and activity are tightly regulated reflecting its importance in cellular homeostasis and response to stress.
Pathways
Spermidine synthase is central to the polyamine biosynthesis pathway which is important for cellular proliferation. In this pathway it acts after ornithine decarboxylase converting putrescine to spermidine and before spermine synthase which produces spermine from spermidine. Spermidine synthase is also involved in the regulation of eukaryotic translation initiation highlighting its influence on protein biosynthesis and cell growth.
Aberrant expression or activity of spermidine synthase has been linked to cancer and neurodegenerative diseases. In cancer elevated levels of polyamines can promote tumor growth and survival.
