ZNF451 also known as COASTER is a protein with a molecular mass of approximately 148 kDa. It functions as a transcription factor by binding to specific DNA sequences regulating the expression of certain genes. ZNF451 achieves this through its zinc finger motifs known for DNA interaction. It contains N-terminal PPP1 binding domain which plays a role in its molecular function. Expression of ZNF451 is noted in various tissues with a higher presence in the heart and skeletal muscle indicating its role in these areas.
Biological function summary
ZNF451 participates in chromatin remodeling an important process in regulating gene expression. It is part of a larger complex that includes components like histone deacetylase. This complex influences chromatin structure by deacetylating histones facilitating transcriptional repression. Through these actions ZNF451 modulates cell cycle regulation and developmental processes. Its involvement in such foundational cellular functions highlights its importance in cell differentiation and growth.
Pathways
ZNF451 plays a significant role in the TGF-beta signaling pathway and the Hippo pathway. In the TGF-beta pathway ZNF451 interacts with SMAD proteins which are critical for transmitting extracellular signals into the cell nucleus affecting gene expression. This interaction modulates processes such as cell proliferation and differentiation. In the Hippo pathway ZNF451 relates to YAP/TAZ proteins helping regulate organ size by controlling cell proliferation and apoptosis. Both pathways underline ZNF451's involvement in maintaining cellular homeostasis and growth.
ZNF451 has connections to cardiovascular diseases and certain muscle disorders. It has been implicated in dilated cardiomyopathy where its dysregulation can affect the structural integrity and function of cardiac muscle. ZNF451’s interaction with cardiac-specific proteins can contribute to the remodeling seen in these conditions. In muscle disorders it influences muscle differentiation and regeneration suggesting that altered ZNF451 function could impact muscle repair. These associations indicate ZNF451's potential as a target for therapeutic interventions in related diseases.
