The PRIM2 protein also known as DNA primase large subunit plays an essential role in DNA synthesis. With a mass of approximately 58 kDa PRIM2 synthesizes short RNA primers needed for DNA polymerase to start replication. This protein is mainly expressed in tissues with high replication activity including the thymus and bone marrow as well as during the S phase of the cell cycle in various cell types. The PRIM2 protein is critical during the initiation of DNA replication binding to single-stranded DNA to form an RNA-DNA hybrid that DNA polymerases extend.
Biological function summary
DNA primase large subunit forms a complex with the smaller subunit PRIM1 creating the DNA primase enzyme. This complex is part of the larger DNA polymerase alpha-primase complex which is fundamental for the initiation of DNA replication. Primase activity ensures that replication begins accurately and efficiently enabling cells to maintain genomic integrity during cell division. By synthesizing RNA primers PRIM2 facilitates the transition from the initiation to the elongation phase of DNA replication.
