The exportin 6 (XPO6) also known as exportin T is a protein involved in nuclear export processes. It has a molecular mass of about 130 kDa. XPO6 is expressed in various tissues including the liver kidney and skeletal muscle where it facilitates the transport of specific proteins and RNA from the nucleus to the cytoplasm. XPO6 functions by binding to its cargo and navigating through the nuclear pore complex effectively regulating intracellular transport.
Biological function summary
Exportin 6 is responsible for the export of profilin-actin complexes from the nucleus playing an important role in maintaining cellular actin equilibrium. It is not part of a multi-protein complex per se but its interaction with cargo molecules involves recognizing nuclear export signals. XPO6's function extends beyond transport influencing cytoskeletal dynamics by regulating actin monomer availability within the cell which is essential for various cellular functions like motility and division.
Pathways
The activities of XPO6 intersect with the Ran GTPase pathway. This pathway facilitates the transport of proteins and RNA between the nucleus and cytoplasm where XPO6 collaborates with Ran GTPase to recognize and bind nuclear export signals of its cargo proteins. XPO6 also interacts with importins and other exportins ensuring balanced nuclear-cytoplasmic transport necessary for cellular homeostasis and response to environmental changes.
Dysregulation of XPO6 has associations with cancer particularly in tumors where actin dynamics are altered. Abnormal expression of XPO6 can disrupt actin balance influencing cell proliferation and migration hallmarks of cancer progression. Additionally XPO6 is connected to neurodegenerative diseases through its impact on cytoskeletal stability. Alterations in XPO6 expression or function can contribute to impaired neuronal transport processes. In both contexts exportin 6 relates to the dysregulation of proteins such as profilin whose stability and localization are critical for normal cellular function.
