PNN also known as Perineuronal Net is a specialized extracellular matrix structure surrounding neurons mainly in the central nervous system. It weighs about 800 kilodaltons. This matrix regulates neural plasticity by influencing cell interactions and neurotransmission. PNN modulates synaptic stabilization and neural circuitry which is essential for maintaining structural integrity within the brain. The PNN expression is notably high in regions like the cortex hippocampus and amygdala supporting roles in cognition and emotional processing.
Biological function summary
The network formed by PNN plays critical roles in neural protection and synaptic plasticity. It acts as a molecular scaffold for key proteins such as aggrecan and tenascin-R forming a complex that can regulate ion homeostasis. PNN confines the activity of enzymes like matrix metalloproteinases maintaining a balanced extracellular matrix environment important for neuron functionality and protecting against oxidative stress. Additionally the stability provided by the PNN components facilitates long-term memory retention and recovery after neural damage.
Pathways
PNN integrates into neural plasticity and learning pathways. It influences the Wnt/β-catenin signaling pathway promoting synaptic plasticity and memory formation. PNN interacts with related proteins such as brevican and neurocan reinforcing inhibitory signals that refine neuronal responses. This connectivity aligns with its role in refining connectivity and maintaining synaptic coverage therefore controlling the duration and specificity of synaptic transmission.
PNN changes often relate to neurodegenerative conditions like Alzheimer's disease and schizophrenia. Altered PNN composition or loss can lead to impaired synaptic regulation and circuitry disruptions. The interaction with amyloid precursor protein often found disrupted in Alzheimer's highlights its influence in disease progression. Furthermore studies suggest a correlation between PNN degradation and dysregulation with proteins like reelin which might contribute to the pathophysiology of schizophrenia indicating a potential target for therapeutic intervention.
