RC3H1 also known as Roquin-1 is a protein with a mass of approximately 125 kDa. It plays a mechanical role in regulating mRNA stability and modulating the immune response by binding to specific mRNA transcripts. RC3H1 contains a unique RING finger and CCCH-type zinc finger domains that assist in recognizing and binding to target RNAs. Expression of RC3H1 occurs in various tissues including lymphoid organs indicating its involvement in immune system regulation.
Biological function summary
RC3H1 contributes to the control of immune homeostasis and inflammation. It acts as part of a complex with other signaling molecules regulating the degradation of target mRNAs by interacting with decay-inducing deadenylation molecules. Its activity ensures the proper regulation of immune responses preventing autoimmunity and excessive inflammation. The protein exerts its effects by targeting mRNAs of cytokines and other immune-related proteins.
Pathways
RC3H1 connects to key immune regulation and inflammatory pathways such as the NF-κB signaling and T-cell receptor signaling pathways. Through these pathways RC3H1 influences the stability and expression of proteins like TNF-alpha and ICOS which are essential for immune responses. These interactions help maintain a balance in immune cell activation and proliferation impacting overall immune function.
RC3H1 has been linked to autoimmunity and chronic inflammatory conditions. Its connection to the development of these diseases involves dysregulation of mRNA degradation leading to alterations in cytokine production and immune cell activation. For instance compromised RC3H1 function associates with systemic lupus erythematosus (SLE) and rheumatoid arthritis (RA) where imbalanced immune responses occur. Studies suggest that RC3H1's interaction with related proteins such as CTLA-4 and other co-stimulatory molecules contributes to the pathological manifestations of these disorders.
