ENAH also known as MENA is a protein involved in cell motility and adhesion. Its molecular mass is approximately 80 kDa. This protein is widely expressed in tissues with high rates of cell movement and remodeling like epithelial tissues. ENAH/MENA belongs to the Ena/VASP protein family which is known for its role in regulating the dynamics of actin filaments essential for cell movement processes such as embryogenesis and wound healing.
Biological function summary
ENAH/MENA influences actin filament elongation and organization participating in the structural integrity of the cell cytoskeleton. It works as part of the actin polymerization complex and cooperates with proteins like profilin and VASP. This cooperation ensures proper cell movement and structure maintenance aiding in processes like axonal guidance and immune cell response.
Pathways
ENAH/MENA impacts the regulation of cell migration pathways including the Rho GTPase signaling pathway. It interacts with key signaling molecules like Rac and Cdc42 which coordinate cytoskeletal reorganization. These pathways are important for directional cell migration which is important in normal developmental processes and tissue repair.
ENAH/MENA has been linked to cancer progression particularly in metastasis and neurological disorders such as amyotrophic lateral sclerosis (ALS). In these contexts its interaction with MET receptor and dysregulated signaling can promote invasive cell behavior and neuronal dysfunction highlighting ENAH/MENA as a potential target for therapeutic intervention.
