SHARP1 also known as DEC2 or bHLHE41 functions as a transcription factor involved in regulation of the circadian rhythm. It belongs to the basic helix-loop-helix (bHLH) family of transcription factors and has a mass of approximately 30 kDa. SHARP1 shows expression in various tissues including the brain liver and skeletal muscle where it contributes to controlling the expression of clock and other genes involved in circadian regulation.
Biological function summary
SHARP1 regulates genes by competing for binding to E-box sequences on DNA often through forming complexes with CLOCK and BMAL1 proteins. This interaction inhibits transcriptional activation altering the rhythmic expression of core clock genes and related metabolic pathways. SHARP1's role extends beyond circadian rhythms influencing processes like cell differentiation and proliferation.

SHARP1 plays roles in circadian and metabolic pathways. In the circadian pathway SHARP1 modulates the feedback loops involving PER and CRY proteins impacting the cycle of gene expression. In metabolic pathways SHARP1 links to glucose metabolism through interaction with PGC-1α a transcription coactivator that regulates energy metabolism.
SHARP1 impacts sleep disorders such as Familial Advanced Sleep Phase Syndrome (FASPS) and some metabolic diseases. The alteration of SHARP1 expression or function can disrupt the circadian rhythms contributing to these conditions. SHARP1 also connects with the PER2 protein in circadian rhythm disorders highlighting its potential role in these pathological states.
