The ENL protein also known as ELL-associated factor 2 is a member of the YEATS domain proteins. With a molecular weight of approximately 54 kDa ENL is expressed widely across various tissues including the brain and testis. Its structural composition allows ENL to interact with chromatin contributing to transcriptional regulation through interactions with histones.
Biological function summary
ENL plays a critical role in the regulation of gene expression by modulating transcription elongation. It exists as part of the super elongation complex (SEC) which is essential in controlling RNA polymerase II pause release during transcription. This function is integral in development and differentiation processes as the ENL protein interacts with other transcription factors.
Pathways
ENL significantly contributes to transcriptional control through its involvement in the transcription regulation pathway. It associates with other members of the SEC such as AF9 and ELL facilitating the transcriptional activation of genes. Additionally ENL interacts with the Wnt signaling pathway influencing cellular proliferation and differentiation.
ENL has a connection to acute leukemias having roles in chromosomal translocations involving the MLL gene. These translocations result in chimeric proteins that cause dysregulated gene expression contributing to leukemogenesis. ENL's interaction with MLL-ENL fusion proteins illustrates its impact on disease progression highlighting its importance as a potential therapeutic target in leukemia research.
