Erbin also known as ERBB2IP (ERBB2 interacting protein) is a scaffolding protein with a mass of approximately 200 kDa. This protein localizes primarily at cell-cell junctions and participates in the stabilization of the basolateral membrane in epithelial cells. Erbin contains a PDZ domain that facilitates its interactions with other proteins allowing it to function effectively in cell signaling. It is widely expressed in various tissues including the brain heart and liver which indicate its importance in different physiological contexts.
Biological function summary
Erbin plays a role in maintaining epithelial cell polarity and adherence. It is part of the leucine-rich repeat and PDZ domain (LAP) protein family and interacts with receptor tyrosine kinases particularly the ERBB family. Furthermore Erbin forms a complex with other proteins to modulate signaling pathways that control cell proliferation and differentiation. Through these interactions Erbin influences key cellular processes that are fundamental for tissue organization and stability.
Pathways
Erbin participates in the ERBB signaling and MAPK/ERK pathways. Through the ERBB signaling pathway Erbin interacts with ERBB2 a receptor tyrosine kinase to regulate downstream signaling that affects cell growth and survival. In the MAPK/ERK pathway Erbin contributes to signal transduction processes important for cellular responses to growth factors. These pathways rely on the precise modulation of signaling by proteins like Erbin to maintain normal cellular function and prevent aberrant cell proliferation.
Erbin has been implicated in cancers and cardiac disorders. Its interaction with ERBB2 links it to breast cancer where dysregulation of this signaling pathway can lead to uncontrolled cell growth. Additionally Erbin is associated with heart failure as changes in its expression or function affect cardiac cell structure and function. In these conditions Erbin's interactions with other proteins can influence disease progression by altering key signaling events important for maintaining normal tissue homeostasis.
