The protein AP2M1 also known as AP-2 complex subunit mu plays an important role in clathrin-mediated endocytosis. It weighs approximately 50 kDa. AP2M1 is mainly expressed in different cell types including neuronal epithelial and fibroblast cells. This protein is part of the adaptor protein (AP) complex family specifically the AP-2 complex which facilitates the recruitment of clathrin and cargo proteins to form clathrin-coated vesicles at the plasma membrane.
Biological function summary
The AP2M1 protein is involved in the sorting and trafficking of membrane proteins and lipids. It functions as part of the AP-2 complex a heterotetrameric assembly composed of two large adaptins a medium chain (mu) adaptin like AP2M1 and a small chain adaptin. This complex helps recognize and bind specific sorting signals found on the cytosolic tails of transmembrane cargo proteins allowing their incorporation into forming vesicles during endocytosis.
Pathways
AP2M1 contributes significantly to the endocytic pathway and the regulation of receptor-mediated signaling. Involvement with the insulin signaling pathway highlights its role in internalizing insulin receptors and modulating insulin sensitivity. AP2M1 interacts with other proteins such as clathrin which influences the assembly of clathrin-coated pits at the cell membrane and also with accessory proteins like dynamin facilitating the fission of vesicles during endocytosis.
Research associates AP2M1 with neurodegenerative diseases such as Alzheimer's disease due to its role in synaptic vesicle recycling and endocytosis affecting neuron function. It also relates to metabolic disorders like diabetes as improper function or regulation of AP2M1 can impact insulin receptor internalization and downstream signaling. Other proteins connected to AP2M1 through these disorders include clathrin and dynamin which are pivotal for maintaining the integrity of cellular transport mechanisms.
