SERPINE2/PN-1 also known as protease nexin-1 (PN1) is a serine protease inhibitor with a molecular mass of approximately 47 kDa. This inhibitory protein predominantly expresses in various tissues including the brain lungs and liver. In the central nervous system it is abundantly present and plays important roles in glial cell function. It acts by binding and inhibiting multiple serine proteases such as thrombin plasmin and trypsin. PN-1 exerts significant mechanical control over these proteases regulating processes like coagulation and fibrinolysis.
Biological function summary
The protein PN-1 is important for maintaining tissue homeostasis and controlling extracellular matrix remodeling. PN-1 does not form part of a larger complex but interacts directly with its target proteases. It shields tissues from excessive proteolytic activity therefore preventing unwarranted degradation of proteins. The protein's inhibitory action is non-specific allowing it to modulate various biological processes from inflammatory responses to tissue recovery through its action across different proteases.
Pathways
PN-1 plays a role in the regulation of coagulation and fibrinolytic pathways. In the coagulation pathway its interaction with thrombin is significant inhibiting thrombin’s activity and therefore controlling blood clot formation. In the fibrinolytic pathway PN-1 modulates the activity of plasmin an essential protease for degrading fibrin clots. By regulating these pathways PN-1 interacts with proteins such as fibrinogen and other regulatory elements within these biological systems.
The function of PN-1 connects closely with cardiovascular diseases and neurodegenerative disorders. In cardiovascular diseases excessive or insufficient activity of PN-1 may contribute to improper blood clot formation linking this protein to conditions like thrombosis. In the context of neurodegenerative disorders altered levels of PN-1 influence the activity of neurological proteases potentially impacting diseases like Alzheimer’s. PN-1 may interact with proteins like α-2-macroglobulin in these disease processes emphasizing its role in pathophysiological conditions.
