RPS16 also known as Ribosomal Protein S16 is a component of the 40S ribosomal subunit with a molecular mass of approximately 16.5 kDa. It plays a role in protein synthesis by joining together with other ribosomal proteins and rRNA to form the ribosome's small subunit. RPS16 is expressed in various tissues throughout the human body reflecting its essential role in maintaining protein synthesis machinery.

This protein's abnormalities can also loosely link to certain cancers as ribosomal dysfunction may lead to uncontrolled cell growth and division through altered expression of proteins like c-Myc which regulates ribosome biogenesis.
