TADA3L also known as Transcriptional Adaptor 3-Like is part of the larger transcriptional coactivator complexes. The protein has a mass of approximately 55 kDa and is expressed in various tissues including liver and kidney. Mechanically TADA3L acts as a transcriptional adaptor interacting with DNA-binding transcription factors and assisting the recruitment of additional coactivators necessary for gene transcription.
Biological function summary
The protein assists in regulating gene expression by serving as a component of the human Ada-Two-A containing (hATAC) complex. The hATAC complex which includes TADA3L plays a role in histone acetylation allowing chromatin to be more accessible for transcriptional machinery. TADA3L's involvement in this complex underlines its role in modulating transcription and influencing cell cycle and proliferation processes.
Pathways
The TADA3L protein integrates into networks influencing transcriptional regulation and chromatin remodeling pathways. It associates with GCN5 and PCAF which are critical for histone acetyl transferase (HAT) activity within these pathways. Through its interaction with these proteins TADA3L contributes to the control of gene expression and is an important part of maintaining homeostasis in cellular function.
Alterations in TADA3L function or expression link to cancer and developmental disorders. Mutations or deregulation of this protein can disrupt normal transcriptional regulation leading to oncogenesis. In cancer pathways TADA3L shows connections with proteins like p53 a widely studied tumor suppressor where its impairment could affect apoptotic and cell cycle arrest pathways contributing to tumor development.
