The target Aflatoxin B1 also called AFB1 and its metabolite Aflatoxin M1 or AFM1 are mycotoxins produced by the molds Aspergillus flavus and Aspergillus parasiticus. AFB1 has a molecular mass of 312.27 g/mol. It is a potent carcinogen and livestock and humans typically encounter it through contaminated food sources. The liver primarily expresses the metabolites of Aflatoxin B1 where it undergoes metabolic conversion to the hydroxylated metabolite AFM1 a less toxic byproduct but still of concern in food safety and regulation.
Biological function summary
Aflatoxin B1 and Aflatoxin M1 disrupt cellular processes and cause DNA damage. AFB1 forms adducts with guanine bases in DNA leading to point mutations. This activity can promote carcinogenesis especially in hepatocytes as they possess enzymes like cytochrome P450 that convert AFB1 to its active epoxide form. These metabolites do not exist as part of a protein structure or complex but have an impact on the cellular environment of the liver potentially affecting detoxification mechanisms.
Pathways
Aflatoxin B1 influences several vital metabolic and signaling pathways within the liver. It links to the cytochrome P450 enzyme system a group of enzymes involved in the detoxification of various xenobiotics and endogenous compounds. This relationship alters the P53 tumor suppressor pathway an important guard against carcinogenesis as mutations in P53 frequently appear in Aflatoxin-related liver cancers.
Exposure to Aflatoxin B1 shows a strong connection with hepatocellular carcinoma a major form of liver cancer. Individuals possessing hepatitis B virus infection display an increased susceptibility to Aflatoxin-induced liver cancer. The relation of AFB1 with the P53 protein is significant where mutations in P53 contribute to Aflatoxin-related carcinogenicity. Additionally exposure to AFB1 and AFM1 correlates with growth impairment in children independent of its carcinogenic effects.
