CDNF also known as Cerebral Dopamine Neurotrophic Factor is a protein that helps in maintaining the proper function of neurons. It has a mass of approximately 18 kDa. This protein is widely expressed in various tissues especially in the brain. CDNF maintains endoplasmic reticulum homeostasis and is involved in the stabilization of dopaminergic neurons. It does this by reducing stress in the endoplasmic reticulum and promoting cell survival mechanisms.
Biological function summary
The protein ensures the survival of neurons by modulating cellular stress pathways. CDNF is not known to be part of a larger protein complex. Instead its activity relies on its own structural configuration to influence stress responses and neuron protection. As a secreted protein it directly interacts with and influences cellular mechanisms within the extracellular space contributing to a healthy neuronal environment.
Pathways
CDNF interacts with at least oxidative stress response pathways and neuroprotection pathways. These pathways underpin its role in counteracting cellular stress related to neurodegeneration. For example CDNF shares a functional connection with proteins like MANF (Mesencephalic Astrocyte-derived Neurotrophic Factor) another neurotrophic factor involved in similar neuroprotective pathways enhancing neuronal survival and recovery from cellular stress.
Neurodegenerative conditions such as Parkinson's disease and Alzheimer's disease are major areas where CDNF shows relevance. In Parkinson's disease CDNF's neuroprotective role helps mitigate the loss of dopaminergic neurons. There is evidence linking CDNF with MANF which also exhibits protective roles in neuronal health. Both proteins aim to promote neuron survival amid progression of neurodegeneration associated with these diseases indicating their potential as therapeutic agents.
