LY75 also known as DEC-205 is a receptor that plays a role in antigen capture and presentation. It has a mass of about 205 kDa. Expressed on the surface of dendritic cells (DC) DEC-205 is considered an important marker for these immune cells. CD103 DC also use the DEC-205 pathway for their functions. Additionally DC-SIGN and its related receptor DC-SIGNR are other DC markers aiding in pathogen recognition. TLR7 part of the Toll-like receptor family is involved in the detection of single-stranded RNA contributing to immune responses.
Biological function summary
LY75/DEC-205 and associated DC markers function in the identification and processing of antigens. They recognize specific ligands and internalize them for processing and presentation to T cells. DEC-205 plays an important role in cross-presentation important for activating cytotoxic T cells. This receptor is part of a larger complex involving other C-type lectin receptors like DC-SIGN that are also present in dendritic cells. These receptors work together to enhance immune responses against pathogens.
Pathways
LY75/DEC-205 and related DC markers are integral to the antigen presentation pathways that activate T lymphocytes. These pathways include the major histocompatibility complex (MHC) class I and II pathways. DEC-205's role in the cross-presentation pathway allows DCs to present antigens to CD8+ T cells often in collaboration with CD103-mediated processes. Related proteins like CD40 and CD80/CD86 are also engaged during T cell priming enhancing immune activation.
LY75/DEC-205 has implications in autoimmunity and infectious diseases. Autoimmune conditions can arise when improper antigen presentation leads to self-reactive T cells. For example faulty presentation in DEC-205 pathways may link to diseases like rheumatoid arthritis. In infectious diseases receptors like DC-SIGN interact with pathogens such as HIV mediating viral entry and immune evasion. Proteins like TLR7 also connect to immune-related disorders by regulating responses to viral infections.
