DENND5A also known as connecdenn 5 serves as a guanine nucleotide exchange factor (GEF). It regulates the activation of small GTPase Rab-GDP to Rab-GTP which is involved in vesicular trafficking processes. The protein has a mass of approximately 127 kDa. DENND5A expression shows presence in many tissues including the brain pancreas and kidney indicating its diverse functional roles across different organ systems.
Biological function summary
DENND5A influences intracellular signaling and membrane trafficking. It does this by activating Rab protein family members essential for vesicle formation movement and fusion. DENND5A participates in endocytic pathways which involves clathrin-mediated endocytosis a pathway critical for material uptake into cells. There is no clear evidence yet whether DENND5A associates within larger protein complexes but its interaction with Rab proteins suggests close collaboration in cellular trafficking tasks.

DENND5A integrates into the membrane trafficking and cellular communication networks. It plays an important role in the endocytosis pathway and syncs with proteins like Rab22A facilitating efficient vesicular transport from the plasma membrane. Another significant pathway is the mannose 6-phosphate receptor pathway where DENND5A influences lysosomal enzyme trafficking vital for cell maintenance.
DENND5A mutations or dysregulations link to neurodevelopmental conditions like epilepsy and intellectual disability. These associations suggest its importance in neural cell signaling. DENND5A itself indirectly interacts with proteins such as RAB18 which are implicated in Warburg Micro syndrome a rare genetic disorder affecting eye and brain development. Through these interactions its role in disease pathogenesis becomes more apparent highlighting the potential for targeted therapeutic strategies.
