TRIM16 also known as Tripartite Motif Containing 16 is a protein involved in several cellular mechanisms. It has a molecular mass of approximately 64 kDa. This protein is part of the TRIM protein family characterized by the presence of a RING finger domain one or two zinc-binding motifs called B-boxes and a coiled-coil region. TRIM16 is expressed in many tissues including the skin lungs and brain. The protein exhibits E3 ubiquitin ligase activity which tags other proteins for degradation.
Biological function summary
This protein plays a role in regulating cell differentiation and proliferation. TRIM16 is not known to form large complexes but its interactions with other proteins allow it to exert its biological effects. It interacts with factors that influence cellular growth pathways contributing to the control of cellular homeostasis. Its functional roles include modulation of specific transcription factors and involvement in the regulation of inflammation.
Pathways
TRIM16 participates in the interferon signaling pathway and the Wnt/β-catenin signaling pathway. Its involvement in the interferon signaling pathway indicates a role in immune response regulation while its activity in the Wnt/β-catenin pathway suggests a function in cellular development and differentiation. TRIM16 interacts with proteins like β-catenin and STAT1 integrating signals from different pathways to maintain normal cellular functions.
Alterations in TRIM16 expression and function associate with cancer and inflammatory diseases. In cancer TRIM16 often acts as a tumor suppressor with its underexpression linked to increased tumorigenic potential. It interacts with proteins such as p53 to carry out these functions. In the context of inflammatory diseases TRIM16 influences cytokine production closely connected with inflammation regulation and interacts with proteins like NF-kB.
