Lysate preparation:
Our lysates are made using RIPA buffer to which we add a protease inhibitor cocktail and phosphatase inhibitor cocktail (ratio: 300:100:10).
This means that the protein of interest is denatured.

REXO2 also referred to as RNase R or Oligoribonuclease is an enzyme with a molecular mass of around 26 kDa. It plays a significant role in the final steps of RNA decay. Mechanically REXO2 functions as an oligoribonuclease catalyzing the degradation of small ribonucleic acids into mononucleotides. This protein is primarily expressed in the mitochondria of the cell where it ensures the proper completion of RNA turnover.
Biological function summary
The enzyme facilitates the maintenance of cellular RNA quality. REXO2 is not a part of any larger complex but its exonuclease activity allows cells to eliminate short RNA fragments which could otherwise accumulate and lead to cellular dysfunction. Efficient removal of these fragments is essential for normal mitochondrial function and energy production within the cell.
Pathways
This enzyme is key to ensuring RNA metabolic balance. It plays a role in the RNA degradation pathway helping to process and clear unnecessary oligoribonucleotides. While REXO2 does not directly interact with other proteins in a complex it works alongside proteins such as PNPase and RNAse II which facilitate different stages of RNA degradation and processing within the cell.
REXO2 has implications in mitochondrial pathologies. Deficiencies or mutations in this enzyme can contribute to mitochondrial disorders which involve proteins such as POLG and Twinkle. Additionally its efficient RNA clearing function ties it to studies on neurodegenerative diseases where aberrant RNA processing can occur. Understanding these connections allows for deeper insight into how deviations in RNA management can translate into disease states.
