Lysate preparation:
Our lysates are made using RIPA buffer to which we add a protease inhibitor cocktail and phosphatase inhibitor cocktail (ratio: 300:100:10).
This means that the protein of interest is denatured.

SECTM1 also known as Secreted and Transmembrane 1 and sometimes as K12 is a protein with a molecular mass of approximately 28 kDa. It expresses both membrane-bound and soluble forms found mainly in epithelial cells and in some types of immune cells such as monocytes. SECTM1 resides in the Golgi apparatus and can be secreted outside the cell indicating its role in cell signaling and communication.
Biological function summary
The protein plays a role in modulating immune responses. SECTM1 interacts with CD7 a cell surface glycoprotein found on T cells and natural killer cells influencing their activation and proliferation. This interaction is important for immune system regulation and SECTM1 can function together with other immune-related proteins but usually operates independently rather than as part of a larger protein complex.
Pathways
SECTM1 is involved in immunological pathways particularly those governing T-cell activation and the broader inflammatory response. Within these pathways SECTM1 connects with proteins like CD7 and is associated with the PI3K/Akt signaling cascade. This linkage influences cell survival and growth bridging innate and adaptive immunity.
SECTM1 has associations with several inflammatory and autoimmune conditions as well as cancer. Abnormal expression levels of SECTM1 have implications in disorders such as rheumatoid arthritis where its interaction with CD7 appears to play a role in disease progression. Similarly in certain cancers altered SECTM1 expression leads to changes in immune cell behavior affecting tumor growth and metastasis.
