Flt3 also known as Flk2 is a receptor tyrosine kinase with a molecular mass of about 160 kDa. It is expressed on the surface of hematopoietic stem and progenitor cells mainly within the bone marrow and early lymphoid organs. Flt3 plays an essential mechanical role in the regulation of hematopoietic cell proliferation differentiation and survival. It can bind the Flt3 ligand to initiate downstream signaling which contributes to various cellular responses.
Biological function summary
Flt3 is involved in the early stages of the hematopoiesis process. It does not act as part of a major complex but collaborates with other cytokine receptors to influence cell fate decisions. The receptor's primary role is the regulation of blood cell formation effectively controlling the development and maintenance of certain lymphoid and myeloid lineages. This role establishes Flt3 as a critical factor in ensuring the proper balance of cell populations during immune responses and hematopoietic recovery.
Pathways
Cell proliferation and survival signaling heavily involve Flt3. It activates the PI3K/Akt and the Ras/MAPK signaling pathways upon ligand binding. These pathways are pivotal for normal cellular responses and are shared with related receptor kinases like c-Kit and PDGFR. Flt3 signaling influences multiple aspects of cellular function and interacts with various proteins to modulate hematopoietic processes.
Flt3 mutations or overexpression are strongly associated with acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL). Mutations often result in uncontrolled cell proliferation due to constant signaling activity. Flt3's role connects with other proto-oncogenes and tumor suppressor proteins involved in hematopoietic malignancies such as c-Kit and RAS. Understanding these interactions helps in designing targeted therapies for leukemia making Flt3 a significant therapeutic target.
