The AF4 protein also known as MLLT2 is a molecular component with an approximate mass of 131 kDa. This protein is typically expressed in hematopoietic tissues where it plays a role in various cellular functions. The AF4 protein acts as a transcriptional activator and is part of the larger AF4/LAF4/FMR2 (ALF) family which plays significant roles in gene regulation. It participates in the transcriptional regulation process by modulating RNA polymerase II activity ensuring proper expression of specific genes important for cell function.
Biological function summary
AF4 influences transcriptional regulation by interacting with the super elongation complex (SEC). The SEC is known for enhancing gene transcription elongation. AF4 contributes to the proper transcription of genes involved in development and differentiation especially affecting the hematopoietic system where it facilitates normal cell differentiation processes. This involvement identifies AF4 as an integral player in maintaining cellular function and identity through its participation in the regulation of genetic transcription.
Pathways
AF4 contributes importantly to the Wnt and Hedgehog signaling pathways. These pathways are essential in controlling cell fate determination proliferation and migration. The protein's interaction with RNA polymerase II places it in a vital position to influence these pathways directly. Within these pathways AF4's functions are related to those of the other proteins like transcription factors TCF/LEF in Wnt signaling and Gli proteins in Hedgehog signaling highlighting its role in gene expression regulation within these cellular processes.
AF4 has been implicated in leukemias particularly in acute lymphoblastic leukemia (ALL) where chromosomal translocations often involve AF4. This translocation disrupts normal gene function leading to the proliferation of leukemic cells. The interaction between AF4 and the MLL protein resulting in the MLL-AF4 fusion protein contributes to oncogenic processes in these leukemias. Moreover abnormalities in AF4 function have been linked to certain neurological disorders though the specific mechanism and associated proteins require further investigation to be thoroughly understood.
