Lysate preparation:
Our lysates are made using RIPA buffer to which we add a protease inhibitor cocktail and phosphatase inhibitor cocktail (ratio: 300:100:10).
This means that the protein of interest is denatured.

Calcium/calmodulin-dependent protein kinase kinase (CaMKK) also known as CaMKKα and CaMKKβ depending on its isoform is an enzyme with a significant role in cellular signaling. It is a serine/threonine-specific protein kinase with a molecular mass of approximately 51–699 kDa depending on the isoform and expression variant. CaMKK is expressed in various tissues but shows high levels in the brain where it influences numerous neural processes. The enzyme works by phosphorylating and activating other kinases such as CaMK1 and CaMKIV thereby regulating a wide range of cellular activities.
Biological function summary
Calcium/calmodulin-dependent kinase kinase participates in calcium-mediated signal transduction pathways. This enzyme is part of a complex with calmodulin (CaM) a calcium-binding messenger protein which modulates its activity in response to intracellular calcium levels. CaMKK acts as a multifunctional kinase that can regulate neuronal functions energy balance and other critical cellular mechanisms.
Pathways
Calcium signaling and AMP-activated protein kinase (AMPK) pathways involve CaMKK. In calcium signaling CaMKK regulates CaMK series kinases which play roles in memory formation and synaptic plasticity. In the AMPK pathway CaMKKβ can activate AMPK a critical regulator of cellular energy homeostasis. CaMKK connects with proteins like CaMK1 CaMKIV and AMPK through these signaling systems helping maintain cellular responses to environmental and metabolic changes.
Calcium/calmodulin-dependent kinase kinase has associations with neurological and metabolic disorders. It influences conditions like Alzheimer's disease through its role in synaptic function and plasticity. Additionally dysregulation of CaMKKβ activity might be connected to obesity as it affects energy balance and glucose metabolism. In these disorders CaMKK interacts with proteins such as CaMKIV in Alzheimer’s influencing cognitive decline and AMPK in obesity impacting metabolic processes.
