MRFAP1 (Morf4 family-associated protein 1) plays a role in chromatin remodeling and transcription regulation. It physically interacts with the tumor suppressor MORF4L1 (also known as Mortality Factor 4-like 1) and other members of the MRG15 complex. MRFAP1 which has a molecular weight of approximately 13.5 kDa is predominantly expressed in the nucleus of cells. Its expression is observed in various tissues but it's notably high in the testes and heart.
Biological function summary
The protein MRFAP1 contributes to the regulation of transcription by serving as a scaffold linking histone acetylation and methylation. This action fosters interactions within the MRN complex which includes MRG15. Through these connections MRFAP1 influences chromatin structure and as a result gene expression. Its role is significant in regulating cellular senescence and differentiation.
Pathways
MRFAP1 is actively involved in the chromatin modification pathway. This pathway coordinates with the action of other proteins such as MRG15 to alter histone acetylation levels impacting transcriptional regulation. Additionally MRFAP1 engages in the DNA damage repair pathway assisting another protein known as RNF8. These interactions anchor MRFAP1 in pivotal cellular functions relating to genetic stability.
MRFAP1 has associations with cancer and certain chromatin-related disorders. Cancer research highlights MRFAP1's contribution to tumorigenesis where changes in its function can disrupt chromatin remodeling leading to unchecked cell proliferation. Furthermore alterations in MRFAP1 can affect its interaction with MORF4L1 influencing cancer progression. The link between MRFAP1 and these proteins provides insights into potential therapeutic targets for treating these conditions.
