Ramucirumab is a monoclonal antibody that targets vascular endothelial growth factor receptor 2 (VEGFR-2). It specifically binds to the extracellular domain of VEGFR-2 inhibiting its ability to interact with its natural ligands. VEGFR-2 has a mass of around 150 kDa and is primarily expressed on endothelial cells which line the blood vessels. Ramucirumab acts by blocking angiogenic signals which are critical for new blood vessel formation.
Biological function summary
VEGFR-2 is an important component in the angiogenesis process which is the formation of new blood vessels from pre-existing ones. It is part of the plasma membrane-spanning receptor tyrosine kinase family. Upon ligand binding VEGFR-2 undergoes dimerization and autophosphorylation activating downstream signaling pathways. These pathways promote endothelial cell proliferation migration and survival. VEGFR-2's role in vessel permeability and inflammation also contributes to its biological significance.
Pathways
VEGFR-2 is integral within the VEGF signaling pathway and endothelial cell signaling pathways. It closely interacts with vascular endothelial growth factor A (VEGF-A) as its main ligand. This interaction triggers a cascade involving phosphatidylinositol-3-kinase (PI3K) and mitogen-activated protein kinases (MAPK) which are important for vascular development and maintenance. VEGFR-2's activity is balanced by its relation to other receptors like VEGFR-1 ensuring proper vessel formation and function.
Ramucirumab has shown effectiveness for conditions such as gastric cancer and colorectal cancer. These diseases often exhibit abnormal angiogenesis which is important for tumor growth and metastasis. VEGFR-2 is upregulated in these cancers making it a target for therapeutic intervention. Ramucirumab's binding to VEGFR-2 prevents VEGF-mediated tumor vascularization and progression. Its relationship with other proteins such as fibroblast growth factor receptors (FGFRs) in cancer highlights the complexity of angiogenic signaling in malignancies.
