DHHC-6 also known as ZDHHC6 is an enzyme that acts mechanically as a palmitoyltransferase attaching palmitate a fatty acid to the cysteine residues of substrate proteins which affects their localization and function. The enzyme weighs approximately 40 kDa. Expression of DHHC-6 occurs in various tissues notably in the liver and pancreas where its role seems significant due to its expression levels. This protein is a member of the zinc-dependent DHHC family and forms part of a larger palmitoylation machinery within cells.
Biological function summary
Palmitoylation modifies proteins to enable their proper folding and trafficking within cellular environments. DHHC-6 is part of various protein complexes that regulate these modifications imparting specific functional properties to target proteins. This regulation impacts cellular processes including signal transduction and protein sorting by ensuring proteins are correctly placed within membranes. Through this function DHHC-6 helps maintain normal cellular operations by stabilizing interactions between proteins and their lipid environments.
Pathways
Expression and activity of this enzyme are integral to processes involving protein acylation and membrane dynamics. DHHC-6 participates predominantly in the endoplasmic reticulum-associated degradation (ERAD) pathway facilitating the management and turnover of proteins within the endoplasmic reticulum. In this pathway DHHC-6 interacts closely with other proteins such as calnexin which is co-involved in ensuring protein folding and quality control occurs effectively.
Aberrant function of DHHC-6 has links to metabolic disorders and neurodegenerative diseases. Research implicates DHHC-6 in the progression of liver diseases particularly steatosis and related conditions partially due to disrupted protein palmitoylation roles in lipid metabolism. Additionally in neurodegenerative diseases such as Alzheimer's improper interaction with proteins like amyloid precursor protein (APP) may contribute to disease pathology highlighting the importance of precise DHHC-6 regulation in disease prevention and management.
