RIN1 also known as Ras and Rab interactor 1 is a protein that plays a role in intracellular signaling. It has a molecular mass of approximately 80 kDa. RIN1 interacts with both Ras and Rab GTPases and acts as a guanine nucleotide exchange factor (GEF) for these proteins. In terms of expression researchers have detected RIN1 in various tissues including the brain and embryonic fibroblasts indicating its involvement in multiple cellular processes.
Biological function summary
RIN1 modulates signaling pathways by influencing the activation state of Ras-related GTPases. It forms part of a larger signaling complex that regulates endocytosis and trafficking of receptors. This activity is essential for receptor-mediated signaling and cellular communication. By affecting these processes RIN1 contributes significantly to the regulation of cytoskeletal dynamics which impacts cell movement and morphology.
Pathways
RIN1 is directly involved in the Ras signaling pathway which is important for cell growth and differentiation. Through its interaction with Ras proteins RIN1 influences downstream effectors like Raf kinases and PI3K which further modulate signaling cascades. RIN1 also participates in the regulation of other pathways linked to receptor internalization partnering with proteins such as Rab5 to coordinate endocytic trafficking.
Disruptions or alterations in RIN1 function have been associated with certain cancers notably lung and breast cancer. These associations highlight its role in abnormal cell proliferation and survival. Moreover RIN1's interactions with Ras proteins correlate with mutations that lead to oncogenic transformations. Understanding RIN1's normal and pathological roles helps reveal potential therapeutic targets for diseases related to Ras-driven signaling dysregulation.
