Key facts
Detection method:Colorimetric,
Sample types:Plasma, Cell culture supernatant, Serum,
Reacts with:Mouse,
Assay type:Sandwich (quantitative),
Sensitivity:= 0.85 ng/mL,
Range:0.819 - 200 ng/mL,
Assay Platform:Pre-coated microplate (12 x 8 well strips)

Product details:
Mouse ARP1 ELISA Kit is designed for the quantitative determination of ARP1 in cell culture supernatants, plasma and serum samples.
Δ Note:
Mouse ARP1 concentration is low in normal serum/plasma and may not be detectable in this assay.
This assay employs an antibody specific for mouse ARP1 coated on a 96-well plate. Standards and samples are pipetted into the wells and ARP1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse ARP1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of ARP1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

The protein ARP1 also known as Actin-Related Protein 1 functions as an important component of the cytoskeleton organization in eukaryotic cells. It is a member of the actin family and shares structural similarities with conventional actin. ARP1 has a mass of approximately 42 kDa. It localizes mostly in the cytoplasm but is also found in the nucleus to some extent. The protein is expressed in a variety of tissues with notable expression in neural tissues.
Biological function summary
ARP1 plays a significant role in cellular processes beyond maintaining structural integrity. It forms part of the dynactin complex which is important for dynein-driven cargo movement along microtubules. This complex includes several other proteins such as p150Glued and dynamitin. By participating in these assemblies ARP1 facilitates vesicular trafficking and is involved in processes like endocytosis and mitosis.
Pathways
ARP1 contributes to vital cellular networks that drive intracellular transport and mitotic spindle orientation. One important pathway where ARP1 acts is the dynein-mediated transport pathway. Within this network ARP1 partners with the protein dynein to facilitate the movement of vesicles and organelles. Another critical pathway is related to mitotic spindle formation where ARP1 influences the positioning and dynamics of the spindle apparatus ensuring accurate chromosome segregation.
ARP1 possesses connections to neurodegenerative diseases and cancer. Alterations in ARP1 function can lead to impaired axonal transport contributing to conditions like Alzheimer's disease. In the context of cancer ARP1 mutations or dysregulation can disrupt normal cellular division and polarity potentially promoting oncogenesis. The association of ARP1 with p150Glued further links it to neurological diseases as mutations in these proteins can exacerbate cellular dysfunction in neurons.
