The MAST3 protein also known as microtubule-associated serine/threonine-protein kinase 3 is a serine/threonine kinase with a molecular mass of approximately 187 kDa. This protein finds expression in various tissues with significant presence in the brain. Functionally MAST3 plays a role in phosphorylation influencing the activity of other proteins through the addition of phosphate groups which is essential for cellular signaling.
Biological function summary
MAST3 protein modulates intracellular signaling pathways by targeting specific substrates involved in neuronal function. This protein is not known to be part of a larger protein complex but interacts with various signaling molecules to execute its functions. In neurons MAST3 influences synaptic plasticity and regulation contributing to proper neuronal communication.
Pathways
MAST3 plays a role in the Wnt signaling pathway which is important for processes like cell proliferation and differentiation. In this pathway MAST3 interacts with proteins such as Dishevelled facilitating discrete phosphorylation events that affect downstream signaling. In addition MAST3 impacts the AKT signaling pathway which is involved in survival and growth of cells by modulating key proteins within the pathway.
MAST3 protein associates with conditions like neurodegenerative diseases and certain types of cancer. In neurodegenerative diseases abnormal MAST3 function can disrupt neuronal signaling contributing to disease progression through altered interactions with tau protein. In cancer aberrant MAST3 expression potentially enhances malignant cell growth and survival by interacting with proteins such as PTEN a well-known tumor suppressor.
