PDGFR beta also known as Platelet-Derived Growth Factor Receptor Beta is a tyrosine kinase receptor with a mass of around 180 kDa. It mainly expresses in cells derived from mesenchymal tissues such as fibroblasts smooth muscle cells and pericytes. PDGFR beta plays an essential role in the cell through binding to its ligand PDGF which induces receptor dimerization and autophosphorylation triggering various downstream signaling cascades. Tropomyosin 3 on the other hand is an actin-binding protein that stabilizes actin filaments and it is found primarily in muscle tissues.
Biological function summary
PDGFR beta's role involves cell proliferation survival and migration making it significant in tissue repair and angiogenesis. It often associates with Tropomyosin 3 in the context of cytoskeleton dynamics particularly influencing structural integrity and function. PDGFR beta is a component of the receptor complex that includes dimerization with other receptor family members like PDGFR alpha making it versatile in its signaling capabilities. Tropomyosin 3's stabilization of actin filaments supports cellular shape and motility which is important during processes such as wound healing and development.
Pathways
PDGFR beta engages in the PI3K-Akt and MAPK signaling pathways two critical pathways regulating cell growth differentiation and survival. Through these pathways PDGFR beta influences numerous biochemical processes by interacting with proteins like Akt and ERK1/2. These interactions further modulate the remodeling of the cytoskeleton wherein Tropomyosin 3 contributes by enhancing actin stability. Both PDGFR beta and Tropomyosin 3 coordinate to influence cellular architecture and responses to extracellular signals.
PDGFR beta has a well-documented association with cancer and fibrosis. Mutations or overexpression of PDGFR beta may drive tumor progression by promoting angiogenesis and cell migration. Tropomyosin 3 is linked to the maintenance of normal cellular structure and its dysregulation is implicated in various muscle disorders. When PDGFR beta is involved in these diseases proteins like PDGF ligands and matrix metalloproteinases frequently act in concert contributing to pathological changes such as excessive tissue remodeling and abnormal cellular proliferation.
