WNK3 also known as With No Lysine [K] kinase 3 is a serine/threonine-protein kinase weighing approximately 203 kDa. Its expression occurs in various tissues with notable presence in the kidney brain and liver. Structurally WNK3 contains several domains that interact with proteins involved in ion transport and signaling pathways. WNK3 executes its mechanical functions by influencing ion transporter activity such as by regulating the sodium-chloride cotransporter (NCC) and the sodium-potassium-chloride cotransporter (NKCC).
Biological function summary
WNK3 plays a critical role in cellular ion homeostasis and modulation of ion transporters in response to osmotic changes. It is part of a complex that regulates ion balance within cells affecting cell volume and blood pressure. Through phosphorylation activities WNK3 can activate or inhibit other kinases impacting cellular responses to various stimuli. These regulatory actions are significant for maintaining proper electrolyte balance and fluid dynamics in the body.
Pathways
WNK3 is involved in regulating the WNK-SPAK/OSR1 pathway which is essential in controlling ion transporter activities in cells. This pathway fine-tunes the activity of the NCC and NKCC transporters. WNK3 is closely connected with other kinases like WNK1 and WNK4 with all playing common roles in blood pressure regulation and ion transport. The coordination between these kinases ensures precise cellular responses to changes in osmolarity and extracellular signals.
WNK3's regulation of ion balance connects it to hypertension and congenital nephrologic disorders. Hypertension links to the dysregulation of electrolyte transport and blood pressure control influenced by WNK3. Furthermore mutations in WNK3 alongside related proteins like WNK1 and NCC can result in altered renal salt handling leading to congenital disorders such as pseudohypoaldosteronism type II. Understanding WNK3's function and interactions offers valuable insights into treatments targeting these conditions.
