The TCF-4/TCF7L2 and TCF-3/TCF7L1 proteins are part of the T-cell factor/lymphoid enhancer-binding factor (TCF/LEF) family. These proteins also known as transcription factor 7 like 2 and transcription factor 7 like 1 function as transcription factors in the Wnt signaling pathway. TCF7L2 and TCF7L1 typically weigh around 65 kDa and 56 kDa respectively. Expression of these proteins occurs in many tissues notably in the colon liver and pancreas. Mechanically they interact with β-catenin to regulate the transcription of Wnt target genes.

TCF-4/TCF7L2 and TCF-3/TCF7L1 play key roles in controlling gene expression essential for cell proliferation differentiation and embryonic development.
