FCHO1 also known as FCH domain only 1 is a protein involved in clathrin-mediated endocytosis playing an important role in the formation of clathrin-coated pits on the plasma membrane. FCHO1 has a molecular mass of approximately 89 kDa. It is expressed in various tissues with higher expression reported in the brain and heart. FCHO1 contains F-BAR domains that facilitate membrane curvature which helps in the recruitment and assembly of clathrin and other scaffold proteins at the cell membrane.
Biological function summary
FCHO1 interacts with other proteins to drive the initial events in the endocytic process. It forms part of a protein complex that includes AP2 and Eps15 which are essential for the sculpting and pinching off of vesicles. This complex modulates the efficiency and specificity of endocytosis a process integral to cellular uptake of nutrients receptors and maintenance of cellular homeostasis. FCHO1 initiates the recruitment of clathrin to the membrane which is an important step in forming vesicles that contain loaded cargo for transport into the cell.
Pathways
FCHO1 is a pivotal player in the clathrin-mediated endocytosis pathway. This pathway is critical for efficient internalization of specific receptors like receptors for nutrients and hormones and involves interaction with proteins such as clathrin and dynamin. These proteins contribute to vesicle formation and pinching off therefore driving the pathway forward. Moreover FCHO1 also interacts with the AP2 adaptor complex which helps in recognizing cargo proteins and assembling them into budding vesicles.
FCHO1 mutations have been linked to neurodevelopmental disorders specifically intellectual disability. Its dysfunction can affect neuronal signaling pathways due to impaired endocytosis leading to altered synaptic transmission. Research also suggests a potential connection between FCHO1 and cancer as improper regulation of endocytosis may contribute to oncogenic pathways. FCHO1's interaction with proteins like clathrin and AP2 further elucidates its role in such disease mechanisms making it a target of interest for therapeutic exploration.
