PspCas13b also known as CRISPR-associated protein 13b is an RNA-guided ribonuclease found in the CRISPR-Cas system of the bacterium Prevotella sp. P5-125. The protein consists of 1291 amino acids giving it a molecular mass of approximately 140 kDa. Researchers have discovered it primarily in bacterial species where it acts as a part of the bacterial immune system against phages. PspCas13b stands out because it exhibits programmable RNA-targeting activity making it effective at recognizing and cleaving viral RNA.
Biological function summary
PspCas13b operates by binding to CRISPR RNA (crRNA) to form an active complex. This complex targets single-stranded RNA with high specificity due to its RNA-guided endonuclease activity. Unlike many other CRISPR proteins PspCas13b does not cut DNA which prevents unwanted off-target DNA cleavage. The protein's RNA-targeting activity makes it a valuable tool for degrading specific RNA sequences in research and it functions independently without forming large multisubunit complexes.
Pathways
PspCas13b fits well into the adaptive immune responses in bacteria where it acts as part of a defense mechanism against RNA viruses and mobile genetic elements. It shows similarities with other RNA-targeting proteins like Cas9 though Cas9 targets DNA instead. Additionally PspCas13b's action is closely aligned with the ribonucleoprotein complexes where crRNA processing proteins initiate the CRISPR immune response allowing precise RNA interference.
Scientists explore PspCas13b’s potential in antiviral therapies and RNA-related diseases given its unique RNA-targeting capacity. There is evidence linking PspCas13b to therapeutic applications in viral infections by identifying and degrading viral RNA without affecting host DNA. Moreover it links to RNA-binding proteins involved in genetic disorders where RNA misregulation occurs. These features highlight the importance of PspCas13b in the development of innovative treatments targeting RNA-based illnesses.
