Phospho-MSK2 / RSK-B (S360) and Total MSK2 / RSK-B ELISA Kit (ab279867) is a very rapid, convenient, and sensitive assay kit that can monitor the activation or function of important biological pathways in human cell lysates. By determining phosphorylated MSK2 / RSK-B protein in your experimental model system, you can verify pathway activation in your cell lysates. You can simultaneously measure numerous different cell lysates without spending excess time and effort in performing a Western Blotting analysis.
This Sandwich ELISA kit is an
in vitro
enzyme-linked immunosorbent assay for the measurement of human phospho-MSK2 / RSK-B and total MSK2 / RSK-B. An anti-pan MSK2 / RSK-B antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and MSK2 / RSK-B present in a sample is bound to the wells by the immobilized antibody and the wells are washed. In select wells, rabbit anti-phospho MSK2 / RSK-B (S360) antibody is added to detect phosphorylated MSK2 / RSK-B. In the remaining wells, biotinylated anti-pan-MSK2 / RSK-B antibody is used to detect pan MSK2 / RSK-B. After washing away unbound antibody, HRP-conjugated anti-rabbit IgG or HRP- conjugated Streptavidin is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MSK2 / RSK-B (S360) or pan MSK2 / RSK-B bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

MSK2 also known as RSK-B is a member of the serine/threonine-protein kinase family and weighs approximately 84 kDa. It plays a role in the phosphorylation of various transcription factors which impact gene expression. You can find MSK2 in the cytoplasm and nucleus of cells with significant expression in neuron tissues and immune cells. In response to external stimuli MSK2 modulates nuclear and cytoplasmic targets influencing cellular responses and adaptations.
Biological function summary
This kinase involves itself in the regulation of transcription by phosphorylating CREB and activating histone acetylation. MSK2 acts in concert with other kinases and proteins forming part of larger signaling complexes that modulate gene expression and chromatin states.
